    Abstract: The present invention provides methods for isolating fetal cells from samples of maternal blood, and for detecting aneuploidies and heritable disorders. Fetal cells are enriched from maternal blood samples using layered immunosorption to specifically bind erythroid cell precursors. In the layered immunosorption method, a substrate such as a microscope slide is coated with a thin layer of erythrocyte membranes. Prior to assay the membranes are activated by binding an antibody against an erythroid cell surface protein. The sample is then added to the substrate and incubated. Nonadsorbed cells are removed by washing, and the bound erythroid cells are permanently attached by fixation and drying. Molecular beacons or other molecular probes are used for differentially detecting fetal and maternal cells. Permeabilizing detergents are used for purifying and detecting fetal cells.
