Publication number: 20100021477Abstract: Described and claimed herein are combinatorial synthetic Fab libraries displayed on a phage pIX protein. The libraries were built on scaffolds representing the most frequently used genes in human antibodies, which were diversified to mirror the variability of natural antibodies. After selection using a diverse panel of proteins, numerous specific and high-affinity Fabs were isolated. By a process called in-line maturation the affinity of some antibodies was improved up to one hundred-fold yielding low pM binders suitable for in vivo use. The grasping portion is provided with a base that is adapted for insertion into a hollow pocket that exists within the trim clip. A pair of retaining arms are maintained at some distance above the base and extend outwardly along the length thereof. The retaining arms each include at least one spring-loaded ball that extends downward from an underside thereof. When the grasping portion is engaged with the trim clip, the trim clip is retained by the presence of the base within the pocket and by a downward force exerted on the trim clip by the spring-loaded balls. In one embodiment, each phage is initially added to the raw sample in concentrations below the detection limit of the final phage detection process. In another embodiment, the parent phages are tagged in such a way that they can be separated from the progeny phage prior to the detection process. Preferred phage detection processes are immunoassay methods utilizing antibodies that bind specifically to each phage. Antibodies can be used that bind to the protein capsid of the phage.
