Abstract: The present disclosure provides methods for producing modified HSCs, wherein the method comprises passing a cell suspension comprising the cell and a payload through a constriction, wherein the constriction deforms the cell, thereby causing a perturbation of the cell such that the payload enters the cell. In some aspects, the payloads are capable of enhancing one or more properties of the HSCs, such that the HSCs are better engrafted within the bone marrow of a subject.
