Publication number: 20230241162Abstract: Bag3 is a multifunctional protein expressed predominantly in the heart, the skeletal muscle, the central nervous system and in many cancers. Although BAG3 was cloned only a decade ago, studies have shown that genetic variants, particularly those that result in haplo-insufficiency, can lead to severe left ventricular dysfunction; however, the full mechanisms responsible have remained obscure. To obviate the influence of heart failure itself on the biology of Bag3, ransgenic mice harboring a single allele knock-out were studied between 8 and 10 weeks of age before any obvious signs of heart failure were evident. The results were surprising and informative. First, it was found that despite a normal phenotype, young Bag3+/? had marked changes in the proteome that were characterized by changes in proteins associated with metabolism and apoptosis. Consistent with this finding, a decrease in the levels of critical proteins charged with maintaining the mitochondrial membrane potential was observed.Type: ApplicationFiled: October 21, 2022Publication date: August 3, 2023Inventors: Arthur M. FELDMAN, Jonathan KIRK

Publication number: 20220241080Abstract: Certain embodiments generally provide an improved tibial base member comprising keel portions that allow one or both cruciate ligaments to be preserved. Other embodiments provide improved lateral and/or medial inserts having a mesial lip that helps relieve and or prevent impingement between the femoral component and the tibial eminence. Other embodiments provide improved femoral components having various chamfers to provide additional clearance with respect to the tibial eminence and posterior cruciate ligament without decreasing bone coverage.Type: ApplicationFiled: April 21, 2022Publication date: August 4, 2022Applicant: Smith & Nephew, Inc.Inventors: Nathaniel Milton Lenz, Richard Michael Smith, Zachary Christopher Wilkinson, Brian William McKinnon, Abraham Biglari Salehi, Jonathan Kirk Nielsen, Michael D. Publication number: 20220168051Abstract: Described here are self-contained surgical navigation systems which include a head-worn display device to be worn by a user during surgery. The system includes a display generator for generating a visual display on the display device, and a sensor suite having at least one tracking camera. The system further includes a support module including: a user-replaceable, modular battery that is removably insertable into a housing of the support module, and a processor unit configured to receive data from the sensor suite and calculate a position and an orientation of at least one marker. The support module is electrically coupled to the head-worn display device to provide power and data to the head-worn display device. The display device and the support module together comprise the entire sensing and computing capability of the system, without requiring external sensors, cameras, computers, or other electrical equipment.Type: ApplicationFiled: February 14, 2022Publication date: June 2, 2022Inventors: Matthew William Ryan, Jonathan Kirk Nielsen

Publication number: 20220133485Abstract: A hinged knee prosthesis comprises a tibial component and a femoral component. The tibial component is configured to attach to a tibia. The tibial component has a bearing surface. The femoral component is configured to hingedly attach to the tibial component and rotate relative to the tibial component. The femoral component comprises a medial condyle and a lateral condyle. The medial and lateral condyles have an eccentric sagittal curvature surface configured to rotate and translate on the bearing surface of the tibial component. A method of rotating a hinged knee through a range of flexion is provided. The method fixedly attaches a femoral component to a tibial component. Axial rotation of the femoral component is induced relative to the tibial component when the hinged knee is flexed.Type: ApplicationFiled: January 12, 2022Publication date: May 5, 2022Inventors: Roger Ryan Dees, JR., Paul Charles Crabtree, JR., Jonathan Kirk Nielsen
