    Abstract: A method for peptide and polypeptide purification that uses a derivatized solid support to capture all of the polypeptides and peptides present in a solution and in a subsequent step quantitatively release all of the polypeptides and peptides bound to the solid support is disclosed. This method provides for quantitative recovery and permits further analysis by any of a variety of analytical methods, including sequencing, chromatography, mass spectrometry and biological assay. The method may further be used for screening peptides and polypeptides that bind to organic molecules such as small molecule drugs. The method does not irreversibly add any unwanted label moieties or otherwise alter the peptides during the release process that could interfere with subsequent processing or analysis steps, in particular, biological assay to assess function in vivo or in vitro in cell-based, whole animal or human testing.

    Abstract: Apparatuses and methods for producing ions from chemical species analyzing chemical species. In one embodiment, an apparatus includes a first ion source assembly that produces a first sample flow and a second ion source assembly that produces a second sample flow. The first and second ion source assemblies operate substantially at atmospheric pressure, and the second ion source assembly is different from the first ion source assembly. The apparatus also includes a separation member positioned at least partially between the first and second ion source assemblies, the separation member being shaped and sized to reduce interference between the first sample flow and the second ion source assembly and between the second sample flow and the first ion source assembly. In one embodiment, the first ion source assembly can include an electrospray ion source assembly and the second ion source assembly can include an atmospheric pressure chemical ionization source assembly.

    Abstract: A high throughput liquid chromotagraphy column assembly for separating a large sample with a selected mass weight and fluid volume. The assembly includes a loading column with a loading chamber with a first inner diameter and a first length. The loading chamber contains a removable cartridge containing a bed packing material on to which sample is loaded and spatially distributed. The cartridge's volume is sufficient to fully load the sample therein but the length of the loading chamber is insufficient to achieve the selected chromatographic separation. A separation column is in fluid communication with the loading column. The separation column has a separation chamber with a diameter smaller than the loading cartridge's inner diameter and a length greater than the cartridge's length. In one embodiment, the cartridge has an inner diameter two or more times greater than the separation chamber's diameter, and the cartridge has a length one-half or less than the length of the separation chamber.
