    Abstract: The disclosure provides methods and related kits, reagents, and systems for selectively deaminating unmethylated cytosine residues in nucleic acid molecules. In some embodiments, the methods and related kits, reagents, and systems are applied for methods of detecting and/or mapping methylated cytosine residues in nucleic acids. The nucleic can be RNA or DNA. Some embodiments include contacting the polynucleic acid with a bacterial cytosine deaminase, for example DddA or SsdA, or functional fragments or derivatives thereof. Representative DddA and SsdA have sequences set forth in SEQ ID NOS:1 and 2, respectively. The bacterial cytosine deaminases of the disclosure are sensitive to methylation and, thus, deaminate only unmethylated cytosines to provide a cytosine to uracil conversion. The conversion can be detected as a C•G-to-T•A transitions in subsequent sequencing analysis.

    Abstract: Systems and techniques for determining a sideslip vector for a vehicle that may have a direction that is different from that of a heading vector for the vehicle. The sideslip vector in a current vehicle state and sideslip vectors in predicted vehicles states may be used to determine paths for a vehicle through an environment and trajectories for controlling the vehicle through the environment. The sideslip vector may be based on a vehicle position that is the center point of the wheelbase of the vehicle and may include lateral velocity, facilitating the control of four-wheel steered vehicle while maintaining the ability to control two-wheel steered vehicles.

    Abstract: A system performs masking of data stored in a database. The system receives change logs from a source database and masks the change logs to generate masked change logs. The system stores the masked change logs in a pre-buffer. If the system determines that the set of masked change logs stored in the pre-buffer corresponds to a complete transaction, the system determines whether any conflicts exist between masked change logs stored in the pre-buffer and masked change logs stored in a main buffer. If the system identifies conflicts, the system applies the masked change logs stored in the main buffer to a masked replica database. This causes the system to move masked change logs corresponding to a complete transaction from the pre-buffer to the main buffer.

The method includes removing the damaged sleeve from a tube; providing a sleeve assembly including a first sleeve with a radially variable end and a retainer; installing the sleeve assembly in the tube so the radially variable end of the first sleeve is received by the support section, the radially variable end being in a radially contracted configuration during installation and being in a radially expanded configuration after the sleeve assembly is installed in the tube; and deforming the retainer from an installation configuration to a retention configuration to retain the radially variable end of the first sleeve in the radially expanded configuration.
