The sonotrode has a recess in which the vessel is held, the recess being formed by facing surfaces of a plurality of protruding portions of the sonotrode separated by slits and arranged around the recess. The sonotrode is coupled to the vessel by dry contact with the vessel without any coupling layers therebetween. The use of such a sonotrode provides the advantages of allowing effective sonication with relatively low energy loss and with a low temperature elevation.

If said analyte is present in the sample an immuno- or chemical reaction occurs in the liquid phase. The separation of bound complex from unbound material is achieved by using said surface where the separation occurs mainly two dimensionally on the surface of the porous surface (e.g. grid). Interestingly, the disclosed surface enables a separation where said complexes are distributed two dimensionally on the porous surface (e.g. grid), whereas unbound materials are distributed three dimensionally. Accordingly, said surface enables both a two and a three dimensional separation.

The closure device further includes a plunger (3), slidably mounted in the bore for the formation of a sealed reagent storage chamber (9) in the space remaining between the closed lid and the plunger. The inner wall of the bore (2) is provided with at least one groove (4), whose depth is so deep as not to be within the reach of the outer diameter of the plunger (3). The groove extends from exterior end of the bore, over such a length as to maintain a gas flow communication between said reagent storage chamber (9) and the exterior end of the cylindrical bore when the plunger (3) is in a partially inserted position.
