  • Patent number: 11884933
Abstract: An elongated or fiber-supported multicellular aggregation of multipotent cells, wherein multipotent cells are arranged in an oblong or longish arrangement with an aspect ratio of a prolate dimension to a perpendicular dimension of at least 2:1, or supported by a fibrous structure, and wherein the aggregate contains cells at different stages of differentiation, and the aggregate contains polar cells; methods of generating such aggregates; methods of developing the aggregates further into tissue organoids and kits for such methods.
    Type: Grant
    Filed: January 11, 2017
Date of Patent: January 30, 2024
Assignee: IMBA—INSTITUT FÜR MOLEKULARE BIOTECHNOLOGIE GMBH
Inventors: Jürgen Knoblich, Madeline A. Lancaster
  • Publication number: 20210095260
Abstract: An in vitro method of producing a bi- or multi-differentiated tissue with at least two tissue types is provided. The method includes the steps of developing a first tissue to a stage of differentiation of interest; developing a second tissue to a stage of differentiation of interest differing from the stage of differentiation of interest of the first tissue; placing the first and second tissue in a vicinity sufficient for fusion by growth, allowing the first and second tissue to grow and fuse to each other, thereby producing a bi- or multi-differentiated tissue including the first and second tissue with different stages of differentiation; bi- or multi-differentiated tissue obtained by such a method; uses of the tissue and kits for performing the method.

  • Publication number: 20200308550
Abstract: A method of generating an artificial 3D tissue culture of a cancer grown in non-cancerous tissue, includes the steps of providing an aggregate of pluripotent stem cells or progenitor cells, culturing and expanding the cells in a 3D biocompatible matrix, wherein the cells are allowed to differentiate to develop the aggregate into a tissue culture of a desired size; wherein at least a portion of the cells are subjected to cancerogenesis by expressing a oncogene and/or by suppressing a tumor suppressor gene during any of the steps or in the tissue culture, and further including the step of allowing the cells with an expressed oncogene or suppressed tumor suppressor to develop into cancerous cells; drug screening methods; oncolytic virus screening methods; a 3D tissue culture; and a kit for performing the inventive methods.
