    Abstract: An apparatus and method for preparing and culturing cells comprises a gas permeable chamber having at least an inlet and an outlet for introducing culture medium and cells. A cell growth substrate placed at the end(s) of the gas permeable chamber for cells anchored or embedded. The present invention discloses a simple but efficient cell culture method by slowly rotating the cell culture apparatus horizontally combining with a partially filled culture media in the gas permeable container in order to be able to expose the carriers and submerge the carriers intermittently, to achieve an optimal cell culture environment through the efficient oxygen transfer, CO2 balance, with sufficient nutrient supply, without vigorous agitation to achieve oxygen transfer. The horizontally rotation movement with the present cell growth apparatus efficient the nutrition, carbon dioxide and oxygen transfer during cultured process for production of cellular protein products, viruses or harvesting the cells.

    Abstract: A single-used matrix sampling device for the single-used, disposable or autoclavble packed-bed bioreactor is provided herein. The present invention includes a tube means and a matrix collecting means. The matrix collecting means configured for collecting the cell culture matrix includes a receptacle and a pulling means connected to the receptacle or the matrix collecting means could be scoop-like in configuration; the tube means is configured for preventing the matrix collecting means from being contaminated while mechanically or pneumatically sampling; and the tube means could be sealed to enclose the sampled matrix collecting means after sampling. A single-used matrix sampling method for a single-used, disposable or autoclavble packed-bed bioreactor is also provided.     Abstract: An external mixing device comprises: arranging an inlet and an outlet on the single-use container; a sterilizable or a single-use pump head connected to the inlet and outlet with a mixer installed inside. Driven by a driving device, the mixer makes the liquid circulate continuously between the pump head and the single-use container. The external mixing device further comprises a sensor chamber installed with a plurality of sensors and extra ports, which are used for introducing solution or gas inside or for sampling and detecting liquid properties. A single-use connector is used to connect the single-use container to the external mixing device, which can be operated aseptically under general environment. The external mixing device is small-sized with the advantage of simplified operation procedures.

The present invention teaches several methods: a method to enhance the integrity of the growth surface by protecting the growth surface in a rigid solid support; a method of use for enhancing the performance of the surface; and a method of modifying a growth surface for eukaryotic and/or prokaryotic cells comprising the steps of increasing surface area by creating porous and 3-D structure, treating a surface to encourage cell attachment, promoting cell growth and proliferation, and disposing the growth surface in any conventional cell cultivating device. The growth surface is able to program degradation and release the cell/tissue mass after the culture is completed.
