    Abstract: The present inventors confirmed that when a brazzein expression recombinant vector for high expression of brazzein in Saccharomyces cerevisiae was prepared and a S. cerevisiae strain Y2805 was transformed with the recombinant vector, the expression level of brazzein was particularly high, thereby completing an optimal expression system for mass-producing brazzein. Further, when the brazzein expression system is cultured under the optimal culture conditions according to the present invention, the amount of brazzein produced is further increased, the purification process is simple, and costs are reduced. Therefore, it is expected that the brazzein expression system according to the present invention can be widely used for mass-producing and commercializing brazzein, which is a sweet protein.

    Abstract: Novel brazzein variants having higher sweetness and the use thereof are provided. The brazzein variants or multi-variants have higher sweetness than a wild-type brazzein protein. Also, a method of preparing the brazzein variants and a food composition for enhancing a sugar content including the same are provided. The brazzein variants or multi-variants have higher sweetness at least twice that of a conventional brazzein protein, and show equivalent properties such as thermal and pH stabilities and high water solubility compared to the conventional brazzein protein. Therefore, a smaller amount of brazzein variants can be used together with a greater amount of other sweeteners such as sucrose, and can be replaced with the other sweeteners. So, the brazzein variants can be widely used as an additive in manufacture of food products.

The method includes expressing human tyrosinase gene, which C-terminal transmembrane domain of human tyrosinase is removed, and separating and purifying the expressed recombinant human tyrosinase. The method according to the present invention have effects on mass-producing the recombinant human tyrosinase having a good enzyme activity without forming insoluble inclusion body, a simple purifying process and a improved purifying yield.
