    Abstract: A self-circularization RNA construct that can be expressed in a DNA vector and simultaneously circularized through a self-targeting and splicing reaction to form a circRNA is disclosed. The circRNA can consist only of a gene of interest which can be a coding, non-coding, or a combination thereof. The gene of interest has the advantage of being able to rapidly express a peptide or protein. The formed circRNA has a circular structure and has a stable and high half-life because 5? and 3? ends are not exposed. Accordingly, functional RNA such as miRNA, anti-miRNA, siRNA, shRNA, aptamer, functional RNA for gene/RNA editing, ADAR (adenosine deaminase acting on the RNA)-recruiting RNA, mRNA vaccine, mRNA therapeutic agent, vaccine adjuvant, and CAR-T mRNA can be produced as a stable circRNA in cells.

    Abstract: A weldable ultrahard insert can include an ultrahard working layer and a weldable metal layer metallically bonded with the working layer. The ultrahard working layer can be any ultrahard material such as PCD, PCBN, metal carbide, ceramic, diamond, or the like. The weldable ultrahard inserts can be formed by charging a reaction vessel with ultrahard materials, including precursors thereof, and placing a weldable metal layer in the reaction vessel with an optional intermediate layer. The assembly can be subjected to a pressure and a temperature sufficient to metallically bond the weldable metal layer to the ultrahard material. The weldable layer is formed as part of the insert in situ which facilitates subsequent welding of the insert to a tool substrate without risking damage to the ultrahard material.

    Abstract: Contoured solid polycrystalline superabrasive material such as twist drill tips and endmill flank segments can be formed by preparing a precursor mold having a plurality of shaped openings each corresponding to a predetermined shape. A specially prepared charge feed can be placed into the shaped openings to form a charged precursor. The charge feed can include a substantially homogeneous mixture of superabrasive source particulates, sintering binder, and optional inorganic bonding medium. A loaded reaction cup-assembly including the charged precursor can be subjected to a pressure, temperature and time sufficient for sintering and formation of the contoured polycrystalline superabrasive material. Reduced finishing steps and increased tailorability of grade and quality of final polycrystalline products can be readily achieved.
