  • Publication number: 20130302782
Abstract: Methods and compositions for the efficient and accurate determination of susceptibility of a hepatitis C virus (HCV) population to an HCV inhibitor are provided. In certain aspects, the methods involve introducing into a cell a patient derived segment, wherein the cell or the patient derived segment comprises an indicator nucleic acid that produces a detectable signal that is dependent on the HCV; measuring the expression of the indicator gene in the presence of varying concentrations of the HCV inhibitor; determining a standard curve of susceptibility; comparing the IC95 fold change, slope, or maximum inhibition percentage of the HCV population to that of a control HCV population, and determining that the HCV population comprises HCV with a reduced susceptibility to the inhibitor when the IC95 fold change value is increased or the slope and/or maximum inhibition percentage is lower for the HCV population as compared to the control population.

  • Publication number: 20130216997
Abstract: Disclosed are methods and systems for the isolation and detection of microbes from a sample. The use of binding agents for isolation of a microbe of interest from a sample are described. In certain embodiments, the methods use ribosome-based and/or bacteriophage-based amplification of the signal in detection of bacteria and other microorganisms. For example, embodiments of the present invention can achieve total amplification of at least 10,000 from a single infected cell.

  • Publication number: 20130217049
Abstract: Disclosed herein are methods and systems for the detection of an analyte of interest by an amplified assay. The method and systems of the invention utilize the high specificity of biomolecules that have been coupled to a solid support to further amplify a signal produced by the interaction of a binding agent with the analyte of interest as a means to detect low levels of the analyte of interest present in a sample. In certain embodiments, for immunochemical (for example, ELISA or RIA) or immunofluorescence assays in which a protein to be identified is bound directly or through capture antibodies to a passivated surface, amplification of ˜10,000-fold can be achieved.

  • Publication number: 20130177926
Abstract: Whole cell, simultaneous target and drug-target assay using differentially labeled antibodies and flow cytometry. First antibody binds to total target and second antibody binds to the drug binding site of the target, thus drug binding will competitively inhibit the second antibody allowing for a competitive inhibition assay of drug-target binding. The assay allows for whole cell analysis and even analysis of mixed populations of cells, yet provides detailed kinetic assessment of drug activity.

  • Publication number: 20130166593
Abstract: Systems, methods, and media for laboratory testing services are disclosed. Medical information can be received from an electronic device. One or more laboratory tests can be recommended based at least in part on the received medical information. For example, if one or more symptoms are received, then one or more laboratory tests may be suggested based at least in part on the received symptoms. As another example, if an order for one or more laboratory tests are received, then one or more additional and/or alternative laboratory tests may be suggested. Suggested laboratory tests can be based on medical symptoms, medical conditions, medical codes, medical history, medications, laboratories, healthcare providers, insurance companies, financial factors, locations, turnaround time, other factors, or a combination thereof. One or more of the suggested laboratory tests can be ordered.
