  • Publication number: 20250144624
Abstract: An automatic system which combines a three-branch sorter with three independently-controlled valves and corresponding collection platforms. The subgroups of droplets are alternatively sorted into three channels followed by being pumped out into PCR tubes on the platforms. Experimentally, this system can realize an accurate collection with a large working range for both pure positive samples and mixed samples with negative ones. This technology effectively dispenses a large number of droplets into small subgroups with quantitative number, which builds the basis for digital droplet microfluidics.

  • Publication number: 20240316557
Abstract: A portable, miniaturized microfluidic droplet-based digital polymerase chain reaction (PCR) device is provided. The device includes a droplet generation layer with a flow-focusing channel and multiple capillary channels, in which the flow-focusing channel has a flow focusing structure to generate droplets from a provided aqueous sample and extract oil, and the multiple capillary channels is configured to perform an oil extraction. An incubation layer includes heating plates, a microfluidic channel, and a container for conducting a PCR thermal cycling program. Controlled by a controller and powered by a power supplier, the PCR thermal cycling program involves maintaining predefined temperatures on each heating plate sequentially. This process ensures the heating of droplets to different temperatures at distinct times, facilitating precise and efficient PCR amplification.

  • Publication number: 20240301517
Abstract: The subject invention pertains to a microfluidic pipeline which enables isolation and analysis of single viruses. Single viruses are encapsulated and manipulated within microfluidic droplets. The subject invention further pertains to the amplification of single viral genomes are amplified and confined within the droplets, followed by extraction and isolation of the single-droplet contents for sequencing analysis.
