    Abstract: Disclosed is an online optimization method for a power consumption policy of a terminal of Internet of things (IoT), including: performing comprehensive scanning on a user terminal and a link, and obtaining a link behavior graph; inputting the link behavior graph into a simulation based optimization (SBO) system to obtain power consumption parameters for minimizing power consumption of the user terminal; performing centralized storage on the link behavior graph and the power consumption parameters; regularly sending, by the user terminal, operation data to a server, and evaluating, by the server, whether a power consumption policy needs to be optimized; and issuing, by the server, a parameter update instruction to complete the upgrading of the power consumption policy of the user terminal. In the present disclosure, the update of the power consumption policy of the user terminal can be conveyed more quickly and more efficiently.

    Abstract: The invention provides an anti-C5 humanized monoclonal antibody with low immunogenicity and low ADCC/CDC function and its application. By modifying the amino acid sequence of the framework region of Eculizumab monoclonal antibody, the immunogenicity was reduced, and the antibody was replaced from IgG2 subtype to IgG1 subtype, and a flexible amino acid sequence was inserted between the CDR3 and CH2 regions of the heavy chain of the IgG1 antibody to reduce the immunogenicity. The purpose of ADCC/CDC function is to improve the stability of the antibody and prolong its half-life. The binding affinity of the monoclonal antibody of the invention to human C5 is similar to that of the original Eculizumab antibody, it can specifically block the complement hemolytic activity of C5 and the production of C5a and can be used for the preparation of C5-targeted paroxysmal nocturnal hemoglobinuria and atypical the drug for the treatment of hemolytic uremic syndrome has excellent clinical therapeutic value.

    Abstract: The present invention provides De-ADCC/CDC activity monoclonal antibodies and their generation method and applications. The present invention modified the antibody sequences of multi-target sites such as PD-L1, PD-1, CTLA4, TNF-?, PCSK9, NGF, CD47 and C5. A flexible amino acid sequence was inserted between the heavy chain CDR3 and CH2 regions of IgG1 antibody to block the physical stress signal transmit from antigen-bound Fab to its Fc region to reduce the ADCC/CDC activity while not significantly increasing the formation of the polymers. The modified antibodies in the present invention and the original antibodies target the same sites, but the antibody subtype or glycosylation state is different. For the modified antibodies in the present invention, the stability is significantly improved or/and immunogenicity is significantly reduced, and the half-life is prolonged. The modified antibodies eliminate the activity of ADCC/CDC.
