    Abstract: Methods for determining copy number for target genomic segments in a genome are described. The method may comprise, for example, obtaining a plurality of sequencing depth signals for a plurality of genomic segments, where a sequencing depth signal corresponds to a number of sequence reads aligned to a locus in a genomic segment, and where the plurality of genomic segments comprises a target genomic segment; determining a first, second, and third statistical moment for the plurality of genomic segments from the plurality of sequencing depth signals; determining a tumor purity and tumor ploidy for the sample from the first, second, and third statistical moment; and determining the copy number of the target genomic segment based on the sequencing depth signals for the target genomic segment, the tumor purity, and the tumor ploidy. The method may further comprise generating a genomic profile for the sample based on the determined copy number.

    Abstract: The present invention belongs to the field of molecular biology and genetics, and in particular relates to a method for editing a bovine gene based on Pro-iCHI. The present invention provides a method for editing a bovine gene based on Pro-iCHI. Protamine is transiently expressed in gene-edited b-haSCs, which are then injected to mature oocytes to obtain reconstructed embryos. Protamine can eliminate abnormal DNA methylation resulting from oocyte intracytoplasmic haSCs injection and enable the nucli to compress into sperm-like structures, and the obtained bovine Pro-iCHI embryos can successfully develop into blastocysts, with a blastocyst rate comparable to that of the embryos obtained by in vitro fertilization. Moreover, in the present invention, a protamine-encoding gene is inserted into a Saccharomyces cerevisiae protein expression vector for transient expression, which ensures that abnormal DNA methylation is erased, without integration into the genome resulting in the insertion of exogenous genes.

    Abstract: A method of erasing an abnormal epigenetic modification in a sheep iCHI embryo and use thereof. A method including the steps of: transferring a recombinant plasmid expressing protamine into a sheep androgenetic haploid embryonic stem cell, then introducing the cell into a mature sheep oocyte, and performing activation and culture to obtain a sheep iCHI embryo without abnormal epigenetic modifications. In the present disclosure, abnormal methylations in iCHI embryos may be erased by transient expression of protamine in o-haSCs. The blastocyst rate of the prepared Pro-iCHI embryos is significantly higher than that of iCHI embryos, and is similar to that of IVF embryos. The resulting embryos can be used for embryo transfer to obtain semi-cloned gene-edited sheep, providing materials for subsequent breeding applications.

    Abstract: Provided is a bovinized CRISPRboCas9 gene editing system, comprising a boCas9 protein and sgRNA. The mucleotide sequence encoding the boCas9 protein is as shown in SEQ ID NO. 1. Further provided are a bovinized CRISPRboCas9 gene editing method, and the use. SEQ ID NO. 1 is obtained by means of optimizing an exogenous Cas9 gene according to the codon preference of bovine cells, so that the transcription and translation efficiency of the Cas9 gene in bovine cells can be significantly improved, and the content of Cas9 protein in the cells is finally increased. Results show that the expression level of the modified bovinized boCas9 protein is twice that of a wild-type control group Cas9, and the target gene cleavage efficiency is at least 6 times that of a wild-type control group.

The present speaker is different from a conventional moving-coil type speaker, and the speaker can be made smaller than a conventional moving-coil type speaker and with respect to high frequency performance, the speaker has higher sound pressure and a better bandwidth.
