Patent number: 12187996Abstract: A device for simulating a function of a tissue includes a first structure, a second structure, and a membrane. The first structure defines a first chamber. The first chamber includes a matrix disposed therein and an opened region. The second structure defines a second chamber. The membrane is located at an interface region between the first chamber and the second chamber. The membrane includes a first side facing toward the first chamber and a second side facing toward the second chamber. Patent number: 12103216Abstract: Systems, devices and methods are provided for fabricating anisotropic polymer materials. According to various embodiments, a fluidic device is employed to distribute a polymer solution and a flow-confining solution in order to generate a layered flow, where the layered flow is formed such that a polymer liquid sheet is sheathed on opposing sides by flow-confining liquid sheets. The fluidic device includes first and second fluid conduits, where the first fluid conduit receives the layered flow. The second fluid conduit has a reduced height relative to the first fluid conduit, such that the layered flow is constricted as it flows through the second fluid conduit. The constriction formed by the second flow conduit causes hydrodynamic focusing, reducing the thickness of the polymer liquid sheet, and inducing molecular alignment and anisotropy within the polymer liquid sheet as it is hardened and as strain is applied during extrusion of the sheet.Type: GrantFiled: March 24, 2021Date of Patent: October 1, 2024Assignees: THE GOVERNING COUNCIL OF THE UNIVERSITY OF TORONTO, BETH ISRAEL DEACONESS MEDICAL CENTER, INC.Inventors: Lian Leng, Stephanie Grainger, Elliot L. Publication number: 20220081676Abstract: The present invention relates to devices including microfluidic devices, e.g. Skin on-Chip (Skin-Chip), for simulating a physiological response to agents and injury, including tattoo injury. In particular, a Skin-Chip is intended for use in replicating the interaction of tattoo ink with skin on a cellular level, including but not limited to mechanisms of wound healing following a tattoo gun and/or tattoo needle induced skin injury; ink particle effects such as pigment retention, pigment distribution and pigment clearance; inflammatory response to foreign particles, i.e. tattoo ink, etc. Further, effects of tattoo inks on simulated microfluidic skin is extended to determine effects of systemic ink exposure upon other organs through use of organ chips, e.g. liver-chips, kidney-chips, Lymph node-chips, etc. In some embodiments, safer ink formulations, e.g. less toxic ink particles, less toxic ink diluents, etc., are contemplated for development and use over currently available tattoo inks and diluents.Type: ApplicationFiled: September 24, 2021Publication date: March 17, 2022Inventors: Lian Leng, Justin Nguyen, Norman Wen, Antonio Varone

Patent number: 9764505Abstract: Methods and devices are disclosed for providing the controlled formation of planar homogeneous or heterogeneous materials using microfluidic devices. In one embodiment, a planar array of microfluidic channels is employed to produce a flowing liquid sheet having heterogeneous structure by spatially and temporally controlling dispensing of polymer liquid from selected microchannels. The resulting liquid sheet is solidified to produce a planar heterogeneous material that may be continuously drawn and/or fed from the plurality of microfluidic channels. The polymer liquid may include a payload that may be selectively incorporated into the heterogeneous structure. In some embodiments, the local material composition is controllable, thereby allowing control over local and bulk material properties, such as the permeability and the elasticity, and of creating materials with directionally dependent properties.Type: GrantFiled: November 23, 2012Date of Patent: September 19, 2017Assignee: THE GOVERNING COUNCIL OF THE UNIVERSITY OF TORONTOInventors: Axel Guenther, Lian Leng, Andrew Wollard, Arianna McAllister, Milica Radisic, Boyang Zhang
