Abstract: An improved nozzle system for a flow cytometer and accompanying methods have been invented for a high efficiency orientation and sorting process of a flat sample and dedicates items such as equine or bovine sperm cells. This improved nozzle system comprises a nozzle with a novel interior surface geometry that can both gently accelerate the cells and can include an elliptical-like, single torsional interior surface element within the nozzle, i.e., a single torsional orientation nozzle. The elliptical-like, single torsional interior surface element may have a laminar flow surface and may produce the simplest flow path for applying minimal forces which act in either an accelerative nature or orienting hydrodynamic forces, namely, the single torsional orientation forces, to orient a flat sample such as animal sperm cells into a proper direction for an analyzing and efficiently sorting process in clinical use, for research and for the animal insemination industry.

Abstract: Improved flow cytometer system particularly adapted to use for sex-selected sperm sorting include enhanced sheath fluid and other strategies which minimize stress on the sperm cells, including a 2.9 percent sodium citrate sheath solution for bovine species and a hepes bovine gamete media for equine species. Improved collection systems and techniques for the process are described so that commercial applications of sperms samples as well as the resulting animals may be achieved.

Abstract: The present invention relates to the isolation and use of an allene oxide synthase enzyme as an antioxidant of lipid peroxides in biological systems. It is based, at least in part, on the discovery that antioxidation is accomplished enzymatically by RPP, a species of allene oxide synthase, in guayule, and on the discovery that the allene oxide synthase RPP disrupts the chain reaction and propagation steps of lipid peroxidation. The present further invention relates to the use of an allene oxide synthase to result in a time-dependent disappearance of conjugated dienes (i.e. lipid hydroperoxides). The allene oxide synthase rapidly converts free or esterified fatty acid peroxides or hydroperoxides into their corresponding epoxides, which, in turn are converted to ketols. The lipid peroxide and hydroperoxide substrates for this enzyme are known to be toxic to biological organisms and can generate additional peroxides by chain propagation reactions.

These systems combine with other techniques, including techniques for enhanced sheath fluid and other strategies which minimize stress on the sperm cells, and, potentially, a 2.9 percent sodium citrate sheath solution for bovine species and a hepes bovine gamete media for equine species. Improved collection systems and techniques for the process are described so that commercial application of sperms samples as well as the resulting animals may be achieved.
