    Abstract: An assay system is provided of great sensitivity and portability where the presence of a specific target in a sample, as well as its concentration (qualification and quantification) is detected by reason of a potential or voltage in a closed circuit, built up a redox reaction. The reaction is produced by binding a capture moiety to an enzymatic redox reaction partner, allowing the capture moiety to bind to any target in the sample, and washing any such bound target. The bound target, if not immobilized, may be immobilized through use of a second capture moiety. Substrate for the enzyme is then added. The action of the enzyme upon the substrate frees electrons, creating a potential across an anode and cathode which may be separated by a membrane.

    Abstract: The present invention provides microdevices for electrochemical detection of target agents in a fluid sample. The microdevices each comprise an elongated microstructure having an inlet for introduction of a fluid sample potentially containing a target agent, and an outlet to allow the sample to flow through the region of the microdevice that allows detection of the target agent. This detection is mediated through hybridization of nucleic acids corresponding to a target agent with device-associated nucleic acids and subsequent binding of an electrochemical detection agent to allow the transfer of an electron to or from the electrode. In specific embodiment, the device comprises: a hollow elongated microstructure with a conductive surface on the internal surface; b) insulating polymer that is uniformly distributed on the conductive surface; c) adapter molecule associated with the insulating polymer, and d) a plurality of associated nucleic acids conjugated to the polymer surface in a specific orientation.
