    Abstract: The present application is directed to a method of sensitizing homologous recombination (HR)-proficient cancer cells to treatment with poly ADP ribose polymerase (PARP) inhibitors. This method involves providing HR-proficient cancer cells and treating the HR-proficient cancer cells with an Ataxia telangiectasia and Rad3-related (ATR) inhibitor under conditions effective to sensitize the HR-proficient cancer cells to treatment with PARP inhibitors, where the treating is carried out without administering PARP inhibitors. This technique can also be used to treat cancer in a subject. Additional methods of treating HR-proficient cancer cells and methods of treating a cancer patient are also disclosed.
