  • Patent number: 10138508
Abstract: The present invention provides a method for measuring a modified nucleobase that increases detection sensitivity for the modified nucleobase in a target nucleic acid. Specifically, the present invention provides a method for measuring a modified nucleobase including: (1) incubating a nucleic acid sample, a capture probe, and a guide probe in a solution; and (2) measuring the modified nucleobase using an antibody against the modified nucleobase in the solution obtained at (1). The present invention also provides a kit for measuring a modified nucleobase including: (I) a guide probe; and (II) a capture probe and/or an antibody against the modified nucleobase.

    Assignee: FUJIREBIO INC.
    Inventors: Tatsuki Matsuno, Mariko Horiike
  • Patent number: 10107802
Abstract: The present invention provides a technique that suppresses a background value of a detection signal to construct an immunoassay system that detects a modified nucleobase. Specifically, the present invention provides a method for measuring a modified nucleobase including incubating a nucleic acid sample, a capture probe, and a solid phase probe in a solution and measuring a modified nucleobase using an antibody against the modified nucleobase in the obtained solution. The present invention also provides a kit for measuring a modified nucleobase including a capture probe, a solid phase probe, and an antibody against a modified nucleobase.

  • Publication number: 20170298412
Abstract: The present invention provides a method and means that can specifically detect a target nucleic acid containing a modified nucleobase. Specifically, the present invention provides a method for measuring a target nucleic acid containing a modified nucleobase, the method comprising: (1) incubating a nucleic acid sample and a heterologous nucleic acid probe having a property of pairing with a modified nucleobase, in a solution; and (2) measuring the modified nucleobase using an antibody against a modified nucleobase having a property of heterologous pairing, in the solution obtained at (1). The present invention also provides a kit for measuring a target nucleic acid containing a modified nucleobase, the kit comprising: (I) a heterologous nucleic acid probe having a property of pairing with a modified nucleobase; and (II) an antibody against a modified nucleobase having a property of heterologous pairing.

  • Publication number: 20160202249
Abstract: An object of the present invention is to provide a technique that suppresses a background value of a detection signal to construct an immunoassay system for detecting a modified nucleobase. Specifically, the present invention provides a method for measuring a modified nucleobase including incubating a nucleic acid sample and a capture probe in a solution and measuring a modified nucleobase using an antibody against the modified nucleobase in the presence of an absorbent polynucleotide in the obtained solution. The present invention also provides a kit for measuring a modified nucleobase including a capture probe, an absorbent polynucleotide, and an antibody against a modified nucleobase.

Abstract: The present invention provides a technique that suppresses a background value of a detection signal to construct an immunoassay system that detects a modified nucleobase. Specifically, the present invention provides a method for measuring a modified nucleobase comprising incubating a nucleic acid sample (e.g., a sample containing a target DNA containing the modified nucleobase) and a heterogeneous nucleic acid probe (e.g., an RNA probe) in a solution and measuring the modified nucleobase using an antibody against the modified nucleobase in the obtained solution.
