  • Publication number: 20250270314
Abstract: The present disclosure relates to the application of antibodies and bispecific antibodies targeting human Cadherin-17 and CD3 as an effective and tissue-specific treatment of CDH17-mediated diseases or disorders such as breast, lung, pancreatic, gastric, and other CDH17 overexpressed cancers. The bispecific antibodies may contain masking domains to minimize systemic toxicity. The masking domains are fused via protease-cleavable linkers to the Fab domains targeting human Cadherin-17 and CD3. The unmasking of the shielded bispecific antibodies occurs predominantly by proteases and enzymes in the tumor microenvironment. The application of such bispecific antibodies thereby minimizes systemic toxicity and expands the therapeutic index.

  • Publication number: 20250230248
Abstract: The disclosure provides antibodies and fragments targeting EGFR, VEGF, PD-L1, or cMET. The disclosure also provides multispecific antibodies that comprise a first variable domain that can bind the epidermal growth factor receptor (EGFR), a second variable domain that can bind cMET, and a third variable domain that can bind PD-L1 or VEGF. The multispecific antibodies are effective in treating cancers and/or other diseases, disorders, and conditions where pathogenesis is mediated by EGFR, VEGF or PD-L1, and cMET.

  • Publication number: 20250215616
Abstract: The present disclosure relates to designs and applications of Variable Heavy Only (VHO) domain regions. The present disclosure provides a VHO library of polynucleotides encoding VHO domains, wherein the VHO domains are designed based on the Vh domain of a human Vh family, and a method of generating the VHO library. The present disclosure also provides a phage library displaying the VHO domains encoded by the VHO library through the use of M13 bacteriophage minor coat proteins such as pIX and pVII and a method of generating the phage library. The present disclosure also provides a method of screening the phage library to identify VHO candidates that are capable of binding a target of interest.

  • Patent number: 6889727
Abstract: An apparatus and method for dispensing protein-containing materials, such as, for example, pastes, gels, lipidic cubic phases, lipidic mesophases, and mixtures of the foregoing into containers, e.g., wells of assay trays. The invention further provides a method for preparing the aforementioned containers for various experiments involving soluble proteins or membrane proteins or both types of proteins. In a preferred embodiment, the apparatus includes (a) a gel dispenser assembly for dispensing small quantities of gels; (b) a pipette probe for dispensing fluids; (c) an actuator for moving the gel dispenser assembly vertically relative to the pipette probe; (d) a liquid-handling robot having a robot arm for moving the pipette probe and the gel dispenser assembly vertically and horizontally relative to containers, e.g.
