  • Publication number: 20250339856
Abstract: The present disclosure relates to a biological fluid processing system and method. The system comprises a fluid processing device comprising at least one fluid path, a pump for providing a pressure in the at least one fluid path, a valve arranged along said fluid path and a first actuator arranged to control the valve to assume a desired opening state of said fluid path. The biological fluid processing system comprises further a processing interface comprising a pump drive for driving the pump of the fluid processing device, and a processing control element comprising a pump control system arranged to control at least the pump drive and a valve control system arranged to control the first actuator. The system is modular. The fluid processing device is comprised in a fluid processing device module having a predetermined fluid processing device configuration. The processing interfaces have a predetermined processing interface configuration.

  • Publication number: 20250066757
Abstract: The present disclosure is directed to a method for separating supercoiled plasmid DNA (pDNA) from a liquid sample. the method comprising the steps of: (a) adding a liquid sample comprising pDNA to a first chromatography material comprising (i) an anion exchange chromatography ligand for binding to pDNA and (ii) a support material allowing convective flow through the first chromatography material, wherein the liquid sample originates from a cell culture harvest and has been subjected to a step of removing RNA before step (a): (b) eluting a liquid sample.

  • Publication number: 20240279637
Abstract: A method, a system and a use of a single use mixing bag for performing alkaline lysis, neutralisation and clarification of bacterial cells for extraction of plasmid DNA. The method comprises providing a suspension of bacterial cells and providing alkaline lysis solution into a single use mixing bag for performing an alkaline lysis of said bacterial cells, wherein said single use mixing bag comprises an integrated mixer. The method comprises furthermore mixing of the content in the single use mixing bag by the integrated mixer during the alkaline lysis and stopping the alkaline lysis by the addition of a neutralisation solution into the single use mixing bag and clarifying the content in the single use mixing bag after the alkaline lysis has been performed by the addition of a flocculate lifting agent into the single use mixing bag, whereby at least two phases are separated inside the single use mixing bag, whereby one of said phases is a neutralised and clarified alkaline solution comprising plasmid DNA.
