    Abstract: The invention relates to novel compositions, combinations and methods relating to compounds which inhibit EZH2 and their uses for treating and/or preventing tumors associated with methyltransferase EZH2. More specifically the invention relates to synergistic bi-therapy compositions for use in a method of treating and/or preventing tumors associated with methyltransferase EZH2, comprising administering to a subject in need, a therapeutically effective amount of the composition, wherein said composition comprises the combination of EZH2 inhibitor and one HMG-COA reductase inhibitor or statin. The present invention also relates to synergistic tri-therapy compositions as well as methods of use thereof for treating and/or preventing specific types of cancers and tumors comprising further administering an effective amount of one or more anti cancer drugs or chemotherapeutic agents in combination with the above bi-therapy compositions.
