  • Publication number: 20220378024
Abstract: A genetically modified non-human animal in which an auxin-inducible degron system controls degradation of a target protein in a living body includes: a chromosome containing a first nucleic acid that encodes a mutant TIR1 family protein having a mutation at an auxin-binding site and having affinity to an auxin analog.

  • Publication number: 20220380782
Abstract: An auxin-inducible degron system kit that controls degradation of a target protein in a non-plant-derived eukaryotic cell, the kit containing a first nucleic acid that encodes a mutant TIR1 family protein having a mutation at an auxin-binding site, an auxin analog that has an affinity to the mutant TIR1 family protein and a second nucleic acid that encodes a degradation tag containing at least a part of an Aux/IAA family protein and having an affinity to a complex of the mutant TIR1 family protein and the auxin analog.

  • Publication number: 20180320186
Abstract: A method is provided for site-specific insertion of foreign DNA into a genome in an animal cell, including (1) a step of constructing a donor plasmid in which DNA of 100 bp to 300 bp having a sequence homologous to an insertion site on a genome is ligated to the upstream and downstream of foreign DNA of 0.1 kbp to 10 kbp, and (2) a step of inserting the foreign DNA of 0.1 kbp to 10 kbp into a target site on the genome by a homology-directed repair-based genome editing method using the obtained donor plasmid.

  • Publication number: 20120115232
Abstract: Provided is a system which can induce the degradation of a protein of interest in a mammalian cell system reliably and stably within a short time. A mammalian cell inducible for protein degradation, the degradation of a protein of interest being induced by an auxin, in which the mammalian cell has both a TIR1 family protein gene from rice and a chimeric gene expressing a protein of interest labeled with a plant Aux/IAA family protein.
