Publication number: 20080227752Abstract: The present invention is directed to methods for inhibition of histone acetyltransferases using glycosaminoglycans. The invention is further directed to methods for treating disorders associated with hyperacetylation by administration of glycosaminoglycans to a patient in need thereof. In one preferred embodiment, the glycosaminoglycan is a heparin or heparan sulfate oligosaccharide. Studies show that removal of sulfate residues from the O-positions of either the uronic acid or the glucosamine did not eliminate the inhibitory activity of heparan sulfate. Since a majority of heparan sulfate binding proteins appear to require O-sulfation, molecules without certain O-sulfations can be used to inhibit HATs while not interacting with most known heparin-binding proteins. In addition, specific sequences of heparin/heparan sulfate can be used to specifically inhibit various HATs.Type: ApplicationFiled: June 28, 2005Publication date: September 18, 2008Applicant: Trustees of Boston UniversityInventors: Matthew A. Nugent, Edward Hsia, Jo Ann Buczek-Thomas

Publication number: 20070141101Abstract: A device comprising an extracellular matrix having an internal pH between 4.0 and 6.0 is discussed. This matrix contains heparin or a heparin-related compound such as heparan sulfate. Preferably the matrix also contains fibronectin or a fragment thereof. The matrix will bind to a protein having a pH dependent binding to heparin such as VEGF, preferably VEGF 121 or VEGF 165. The device will release the protein as the pH increases to physiological pH, such as 7.0 to 7.5. The device can be used to deliver a drug to a specific site. For example, with VEGF to a site in need of angiogenesis.Type: ApplicationFiled: April 12, 2004Publication date: June 21, 2007Applicant: The Trustees of Boston UniversityInventors: Matthew Nugent, Adrienne Georges

Publication number: 20060222886Abstract: One embodiment of the present invention provides an organic light emitting device comprising an anode, a cathode, and an organic layer disposed between the anode and the cathode, wherein the organic layer comprises an arylpyrene compound. In another embodiment, the present invention also provides organic light emitting devices comprising an anode, a cathode, and an organic layer disposed between the anode and the cathode, wherein the organic layer comprises an arylpyrene compound doped in an anthracene host, preferably ADN. Another embodiment of the present invention provides specific arylpyrene compounds.Type: ApplicationFiled: April 4, 2005Publication date: October 5, 2006Inventors: Raymond Kwong, Matthew Nugent

Patent number: 5766584Abstract: A composition and method are provided for inhibition of vascular smooth muscle cell proliferation following injury to the endothelial cell lining of a blood vessel such as resulting from angioplasty, vascular bypass surgery or organ transplantation. The composition is a matrix such as a biodegradable hydrogel made of a synthetic polymer, protein or polysaccharide seeded with vascular endothelial cells which can be xenografts, allografts or autografts, or genetically engineered cells. Attachment of cells to the matrix can be enhanced by coating with collagen, laminin, fibronectin, fibrin, basement membrane components or attachment peptides. Biologically active compounds such as anti-inflammatory agents may also be contained in the matrix. In the method, the matrix containing endothelial cells is implanted in a patient at a site adjacent the injury such as by wrapping the matrix around the blood vessel.Type: GrantFiled: June 2, 1995Date of Patent: June 16, 1998Assignee: Massachusetts Institute of TechnologyInventors: Elazer R.
