    Abstract: The present disclosure is directed to novel polynucleotide sequences for use in Nannochloropsis gaditana. The novel polynucleotide sequences include control sequences and coding sequences. Also disclosed are novel gene expression constructs wherein N. gaditana promoters/control regions are operatively linked to N. gaditana or non-N. gaditana coding sequences. These novel polynucleotide sequences and expression constructs can be introduced into N. gaditana and can recombine into the N. gaditana genome. Expression from these polynucleotide sequences and expression constructs can enhance N. gaditana biomass and/or lipid biosynthesis. Also disclosed are methods for modifying N. gaditana, for example by stably transforming N. gaditana with nucleic acid sequences, growing the modified N. gaditana, and obtaining biomass and biofuels from the modified N. gaditana.

    Abstract: A system for measuring hydrogen (H2) and oxygen (O2) including a sample system, a signal processing and system control device, at least one polarograph device and a computing device equipped with software for environmental and biological data analysis incorporating both user-defined and environmental data acquired by the signal processing and system control device. The system measures gas level or exchange (production and/or consumption) in the field or laboratory. In some embodiments, the system uses two Clark-type polarograph devices oppositely polarized with gas-specific polarographic circuits, wherein one polarograph device measures H2 gas and the other measures O2 gas. In some embodiments, the sample system includes an environmentally-controlled sample housing for accepting the polarograph devices and holding a test sample in a reservoir. The system measures H2 or O2 exchange in microbial samples, including microbial phototrophs which couple light harvesting to H2 and/or O2 exchange.

    Abstract: Provided herein are methods for measuring H2 production by a hydrogenase. Also provided are methods for evolving a hydrogenase or hydrogen-sensing system by comparing the level of production of H2 by the hydrogenase to the level Of H2 produced by a wild-type hydrogenase cultured under the same conditions and selecting a microorganism with increased H2 production over the H2 production by a microorganism having a wild-type hydrogenase. Further provided herein are microorganisms and plasmids comprising a hupSL promoter coupled with a reporter gene.
