Publication number: 20240240160Abstract: The present disclosure provides a poly nucleotide comprising: (a) a nucleic acid sequence encoding an olivetol synthase (OLS): and (b) a heterologous regulatory element operably linked to the nucleic acid sequence. The present disclosure further relates to an engineered cell comprising an olivetol synthase (OLS). Also provided are a cell extract or cell culture medium or a composition comprising 3.5.7- trioxododecanoyl-CoA, olivetol, olivetolic acid, a cannabinoid, and/or an isomer, analog, or derivative thereof, and a method of making 3.5.7-trioxododecanoyl-CoA, olivetol, olivetolic acid, a cannabinoid, and/or an isomer, analog, or derivative thereof. Also provided is a cannabinoid produced by the engineered cell, isolated from the cell extract or cell culture medium, and/or made by the method described herein. The present disclosure also provides a non-natural olivetol synthase (OLS).Type: ApplicationFiled: May 27, 2022Publication date: July 18, 2024Inventors: Andreas W. Patent number: 11601525Abstract: Methods and apparatus for retrieving and delivering content in a network. In one embodiment, unique data codes representative of content are generated by a network entity. The data codes are distributed to viewers either via a physical medium or on a display of similar content. The user device requests the content from the network by reading the data code. The requested content is delivered directly to the requesting device or provided to another device identified by the requesting device or user. In another alternative, the unique data code may be representative of the user or subscriber himself. This type of data code may be generated by the network or the device, and enables the user to access content he is entitled to via another device or terminal by delivery of information contained in the data code (or the code itself) to the network.Type: GrantFiled: January 28, 2014Date of Patent: March 7, 2023Assignee: TIME WARNER CABLE ENTERPRISES LLCInventors: Jennifer Chun, Michael Angus, Julie Melendez, Mark Wang, James Gutierrez

Publication number: 20230051743Abstract: Methods and apparatus for selectively enforcing required insertion or viewing of secondary content, such as advertisements or promotions, in a content distribution network. In one embodiment, the network is a managed network (e.g., cable television, satellite, or the like), and the method includes identifying secondary content with a particular attribute or property (e.g., an advertisement that a user has watched before), and selectively enabling one or more “trick mode” functions relating to that secondary content. In one implementation, audio codes or watermarks are inserted into the secondary content and detected when the content is accessed or rendered; if the detected content matches a database or listing of previously played content for that user or premises, the trick mode functionality (e.g., FF or “skip”) is enabled, thereby allowing the user to bypass the content.Type: ApplicationFiled: November 1, 2022Publication date: February 16, 2023Inventors: Peter Stern, Michael L. LaJoie, Michael Angus

Publication number: 20220333142Abstract: Disclosed are trans-enoyl CoA reductase (TER) enzymes and nucleic acids encoding them. In some cases, the TER enzymes are non-natural, engineered trans-enoyl CoA reductase. TER enzymes were shown to catalyse the conversion of 5-carboxy-2-pentenoyl-CoA into adipyl-CoA for improved adipate production and the conversion of crotonyl-CoA into 6-aminocaproate. The enzymes can be used in biosynthetic methods and engineered microorganisms that enhance or improve the biosynthesis of 6-aminocaproate, hexamethylenediamine, caproic acid, caprolactone, or caprolactam. The engineered microorganisms include exogenous TER and in some cases engineered TER.Type: ApplicationFiled: April 24, 2020Publication date: October 20, 2022Inventors: Amit M. Shah, Harish Nagarajan, Michael Angus Noble

Publication number: 20220315969Abstract: Described herein are olivetolic acid cyclases (OAC) including non-natural variants capable of forming a 2,4-dihydroxy-6-alkylbenzoic acid from a 3,5,7-trioxoacyl-CoA or a 3,5,7-trioxocarboxylate substrate. In some examples, the non-natural OAC is capable of forming a 2,4-dihydroxy-6-alkylbenzoic acid from a 3,5,7-trioxoacyl-CoA or a 3,5,7-trioxocarboxylate substrate at a greater rate. In some examples, the non-natural OAC has a higher affinity for a 3,5,7-trioxoacyl-CoA or a 3,5,7-trioxocarboxylate substrate, as compared to the wild type OAC. The non-natural OAC can be used with olivetol synthase (OLS) to form the 2,4-dihydroxy-6-alkylbenzoic acid from malonyl-CoA and acyl-CoA through to a 3,5,7-trioxoacyl-CoAintermediate. The non-natural OAC (and OLS) can be expressed in an engineered cell having a pathway to form cannabinoids, which include CBGA, its analogs and derivatives. CBGA can be used for the preparation of cannabigerol (CBG), which can be used in therapeutic compositions.Type: ApplicationFiled: June 5, 2020Publication date: October 6, 2022Inventors: Michael Angus Noble, Russell Scott Komor

Publication number: 20170134772Abstract: A content delivery resource in a cable network receives a request for specified content. The content delivery resource retrieves profile information assigned to the subscriber domain. The profile information indicates multiple different playback formats assigned to the subscriber domain. To service the request, the content delivery resource utilizes the profile information associated with the subscriber domain to select versions of the specified content encoded in accordance with the multiple different playback formats. The content delivery resource then initiates transmission of the versions of the specified content in the multiple different playback formats to the subscriber domain for playback on multiple types of playback devices.Type: ApplicationFiled: January 27, 2017Publication date: May 11, 2017Inventors: James S. Manchester, Michael Angus, Wilfred Jaime Miles
