  • Patent number: 7718400
Abstract: DNA sequences comprising nucleic acids encoding fusion proteins comprising an Fc portion of an antibody attached at the N-terminus of an OB protein moiety, vectors comprising such DNA sequences, host cells comprising such vectors or DNA sequences, and processes for preparing such fusion proteins, and pharmaceutical compositions comprising such fusion proteins, are described.
    Type: Grant

Date of Patent: May 18, 2010
    Assignee: Amylin Pharmaceuticals, Inc.
Inventors: Mary Ann Pelleymounter, Christopher Francis Toombs, Michael Benjamin Mann
  • Patent number: 7524940
Abstract: The present invention provides methods and compositions for treating excess weight by administering OB protein in a form for constant supply, at a dosage of less than or equal to about 1 mg protein/kg body weight/day. Compositions and methods used for production of recombinant murine and human OB protein are also provided. Compositions and methods for preparing recombinant murine methionyl OB protein and recombinant human methionyl OB protein, including DNA sequences, vectors, host cells, methods of fermentation, and methods of purification are provided herein.

Date of Patent: April 28, 2009
    Assignee: Amgen, Inc.
Inventors: Mary Ann Pelleymounter, Randy Ira Hecht, Michael Benjamin Mann
  • Publication number: 20090082275
Abstract: The present invention provides methods and compositions for, for example, effecting weight loss and treating excess weight by administering OB protein, or a derivative thereof, in a form for constant supply, for example, continuous administration, at a dosage of less than or equal to about 1 mg protein/kg body weight/day.
    Type: Application
    Filed: November 12, 2008
    Publication date: March 26, 2009
    Applicant: AMGEN, INC.
Inventors: Mary Ann Pelleymounter, Randy Ira Hecht, Michael Benjamin Mann
  • Patent number: 7208577
Abstract: The present invention provides methods of creating and using OB protein compositions with an antibody constant region or portion thereof fused to an OB protein. The fusion protein is created by attaching the polyamino acids to the OB protein moiety. The fusion proteins can then be used for various therapeutic uses.

  • Publication number: 20030203837
Abstract: The present invention teaches compositions and methods relating to leptin fusion proteins comprising an antibody constant regions optionally attached to the N-terminus of the OB proteins. The compositions of the present invention may be used for treating obesity, diabetes, increasing lean muscle mass or for control of body composition.

  • Publication number: 20030040467
Abstract: The present invention stems from the observation that administration of OB protein to non-obese as well as obese animals results in an increase of lean tissue mass. Thus, OB protein has the capacity to act, in addition to acting as a weight reducing agent, as an agent affecting lean tissue mass. As such, numerous lean tissue-mass increasing therapies are contemplated, even for patients who would not necessarily benefit from weight reduction. Thus, one aspect of the present invention is the use of OB protein (or analogs or derivatives thereof) for increasing lean tissue mass.
