Patent number: 11846633Abstract: Seroprotection analysis systems, kits, and techniques are described for testing oral fluid for the presence of protective levels of antibodies of interest. In one aspect, the presence of protective levels of target antibodies indicates a test subject has achieved a target seroprotection level, such as but not limited to a target seroprotection level following vaccination. A collection kit can include a swab that enables collection of oral fluid containing antibodies from both dentulous and edentulous individuals, a container that can be pre-filled with an extraction solution, and a nozzle that can be coupled to the container to form a fluid-tight device. A user can use the nozzle to seal the swab in the container and to compress the swab within the container.Type: GrantFiled: December 23, 2019Date of Patent: December 19, 2023Assignees: Becton, Dickinson and Company, University of Maryland, BaltimoreInventors: Michael D. Publication number: 20210148906Abstract: Lateral flow assay devices, systems, and methods described herein separate components of a fluid sample, including small volume, undiluted, unprocessed samples. In one aspect, components are retained in a separation membrane that is spatially above and in fluid communication with a conjugate pad of a lateral flow assay. Devices, systems, and methods described herein can retain particles from a fluid sample that obstruct flow of the fluid sample through the conjugate pad to a detection zone and/or interfere with detection of an analyte of interest in the detection zone.Type: ApplicationFiled: January 27, 2021Publication date: May 20, 2021Inventors: Michael D. Fiechtner, Francis R. Go, Scott Castanon

Patent number: 6893822Abstract: The present invention relates to conjugates of synthetic binding units and nucleic acids. The present invention also relates to methods for sorting and immobilizing nucleic acids on support materials using such conjugates by specific molecular addressing of the nucleic acids mediated by the synthetic binding systems. Particularly, the present invention also relates to novel methods of utilizing conjugates of synthetic binding units and nucleic acids to in active electronic array systems to produce novel array constructs from the conjugates, and the use of such constructs in various nucleic acid assay formats. In addition, the present invention relates to various novel forms of such conjugates, improved methods of making solid phase synthesized conjugates, and improved methods of conjugating pre-synthesized synthetic binding units and nucleic acids.Type: GrantFiled: July 19, 2001Date of Patent: May 17, 2005Assignee: Nanogen Recognomics GmbHInventors: Markus Schweitzer, Richard R. Publication number: 20010049111Abstract: This invention relates to devices and methods for carrying out multi-step and multiplex immunoaffinity binding reactions in microscopic formats. In particular, these devices and methods allow the user to rapidly carry out multiple immunoassays in the same sample volume, and to rapidly resolve the results of those immunoassays in an electronically assisted format. The assays may be further multiplexed in that several samples may be analyzed and visualized on the same microelectronic array.Type: ApplicationFiled: February 14, 2001Publication date: December 6, 2001Inventors: Norbert Windhab, Michael J. Heller, Richard R. Anderson, Michael D. Fiechtner, Tina S. Nova, Markus Schweitzer, Alfred R. Sundquist, Christoph Brucher, Jill M. Orwick, Jochen Muller, Stefan Raddatz, Donald E. Patent number: 5686316Abstract: The invention is a rapid, continuous test for glycated hemoglobin using a non-equilibrium affinity binding method. Agarose beads derivatized with 3-aminophenylboronic acid specifically bind glycated hemoglobin. This solid phase is incorporated into a sample processor card, modified to mix and to separate the test solution from the solid phase prior to absorbance readings. Two absorbance readings are made on the test solution, one immediately after mixing the reagent/diluent with the specimen, and one after a significant amount of binding has occurred. A linear correlation between total glycated hemoglobin and hemoglobin A.sub.1c permits standardization and reporting of units equivalent to % hemoglobin A.sub.1c. Stable glycated hemoglobin solutions for use as standards in the assay, and a method for preparing the standards are also disclosed.Type: GrantFiled: April 28, 1995Date of Patent: November 11, 1997Assignee: Abbott LaboratoriesInventors: Michael D. Fiechtner, John M. Ramp, Barbara J. England, Mary J. Annino

Patent number: 4970074Abstract: The present invention provides novel fluorescein derivative compounds having fluorescence spectrum and quantum yield characteristics similar to those of fluorescein. The compounds are readily synthesized and purified and are readily soluble in water at self-quenching concentrations. Significantly, due to the presence of polar polyhydroxy group substituents and the absence of metal-chelating groups, these fluorescein derivatives are susceptible to minimal leakage across liposome membranes and have fluorescence characteristics minimally sensitive to the presence of metal ions. Compounds of the invention are thus exceptionally suitable for use in the development of highly storage stable liposome preparations to be employed in immunolytic assays involving human body fluid samples.Type: GrantFiled: July 24, 1989Date of Patent: November 13, 1990Assignee: Abbott LaboratoriesInventors: Michael D. Fiechtner, Christopher Bieniarz, Mohamed Shipchandler, Maciej Adamczyk
