    Abstract: Provided herein are improved methods for producing a recombinant polypeptide or virus particle. In some embodiments, a method for producing a recombinant polypeptide or virus particle disclosed herein comprises providing a cell culture comprising cells capable of producing the recombinant polypeptide or virus particle and dextran sulfate, and transfecting the cells by adding to the culture a composition comprising one or more polynucleotides and a transfection reagent. In some embodiments, the recombinant virus particle is a recombinant AAV (rAAV) particle.

In some embodiments, a method for producing recombinant AAV (rAAV) particles provided herein comprises culturing cells capable of producing rAAV particles in the presence of a histone deacetylase (HDAC) inhibitor. In some embodiments, a method for producing recombinant AAV (rAAV) particles provided herein comprises culturing cells capable of producing rAAV particles in the presence of a histone deacetylase (HDAC) inhibitor and increased amount of a sodium salt.
