    Abstract: Provided are compositions and methods for modifying DNA substrates. The compositions include modified I-F3 proteins for use in a CRISPR systems to modify a DNA substrate. The modified proteins include I-F3 TnsC, TniQ, TnsA, TnsB and fusion proteins containing TnsA and TnsB, Cas8, Cas5, Cas7, and Cas6 modified proteins. The CRISPR systems include a guide RNA. Protein modifications provide for a higher transposition frequency than unmodified I-F3 CRISPR systems.
