    Abstract: A direct chemical lysis composition includes an assay compatible buffer composition and an assay compatible surfactant. When combined with a specimen storage composition, such compositions prevent undesired modifications to nucleic acid and proteins lysed from cells in the biological sample. Assays of samples from such compositions do not require expensive and time-consuming steps such as centrifugation and prolonged high temperature processing. The direct chemical lysis composition of the present invention permits direct nucleic acid extraction from the cells in the biological sample without the need to decant off the transport media or otherwise exchange the transport media with assay compatible buffers. There is no need to combine the sample with proteinase K or another enzyme to extract nucleic acids from the cells. A method for lysing cells to obtain target nucleic acid for assay and a kit for combining the direct chemical lysis composition with a sample are also contemplated.

    Abstract: A protection circuit provides transient protection to a solid state circuit of a line interface for wired telecommunications media at a voltage level below the power supply of the solid state circuit and prevents transient currents from entering the power supply distribution where they might cause damage to the protected interface and other circuits. The protection circuit uses a MOSFET, the gate of which is provided with a bias derived from a power supply of the solid state circuit being protected. The bias causes the MOSFET to be always on such that the MOSFET provides an immediate and high transconductance path to ground for transient currents from the line interface.

Tryptophan and pyruvate are added to a bioreactor to form a mixture comprising monatin and a plurality of intermediates via a multi-step equilibrium pathway. The methods and systems include operating the bioreactor such that a temperature of the mixture in the bioreactor is less than 25 degrees Celsius, resulting in an increased production of monatin. In some embodiments, the temperature of the mixture in the bioreactor is between about 5 degrees Celsius and about 23 degrees Celsius; in other embodiments, the temperature is between about 10 degrees Celsius and about 18 degrees Celsius.

    Abstract: A filtration system for a particulate-laden stream of gas utilizes a group of fabric bags within the upper chamber of a housing as the means by which particulate matter is filtered out of the gas. The gas inlet into a lower chamber of the housing is provided with an inlet guide that directs the incoming gas against the lower side of a porous diffuser located below the group of bags and spanning at least most of the separation zone defined by the bags. Passages through the diffuser are configured to present an array of nozzles so that the gas is subjected to a pressure drop across the diffuser before flowing to the bags. The nozzle effect causes the particulate-laden gas to spread out evenly on the lower, upstream side of the diffuser and pass through the diffuser at substantially equal velocity at all passages, thereby smoothing the flow and distributing it evenly to all bags.
