Publication number: 20240238441Abstract: The present invention is based on the surprising finding that Wnt7a, and other Wnt family members, are trafficked to extracellular vesicles (EVs) via interactions with coatomer proteins. Extracellular vesicle signal peptides (ESPs), each comprising at least one key Coatomer binding motif (CBM), are described, and these mediate EV trafficking of Wnt family members. The ESPs may be used to target other proteins for display on EVs. Herein is described an EV comprising: COPI, and a recombinant EV-directed polypeptide comprising: a cargo polypeptide, and an ESP comprising a CBM, wherein the cargo polypeptide is tethered to an external surface of the EV via the coatomer binding motif. Also described are recombinant EV-directed polypeptides comprising an ESP and a cargo. Additionally described are recombinant skeletal muscle-targeted EVs comprising a pay load polypeptide; and recombinant Wnts having mutated CBM or ESPs, which are less trafficked to EVs for production/recovery of free Wnts.Type: ApplicationFiled: May 3, 2022Publication date: July 18, 2024Inventors: Michael A. RUDNICKI, Uxia GURRIARAN-RODRIGUEZ, David Alexander DATZKIW

Patent number: 10828346Abstract: There are provided compositions and methods for modulating stem cell division decisions, in particular, division symmetry. It has been demonstrated that wnt7a acts through frizzled-7 receptor expressed on the surface of adult stem cells, e.g. satellite stem cells, to activate the planar cell polarity (PCP) pathway, thereby promoting symmetrical expansion of stem cells. The compositions and methods of the invention are useful, for example, in modulating stem cell division symmetry in vitro and in vivo, in replenishing and expanding the stem cell pool, and in promoting the formation, maintenance, repair and regeneration of tissue.Type: GrantFiled: August 10, 2018Date of Patent: November 10, 2020Assignee: Ottawa Hospital Research InstituteInventors: Michael A. Patent number: 9815413Abstract: A cargo carrier device supports cargo thereon for movement relative to a truck box between a loading position substantially at ground level rearward of the truck box and a transport position above the vehicle. The device includes a cargo frame defining a planar rack for supporting cargo thereon above a passenger cab of the truck in the transport position. A cargo supporting crossbar can be selectively positioned above the planar rack which is adapted to suspend cargo therefrom. Publication number: 20160009373Abstract: A propeller assembly has a main hub and a propeller hub circumferentially movable relative to the main hub about a main axis of the assembly. Circumferentially spaced propeller blades on the propeller hub are i) circumferentially movable together with the propeller hub relative to the main hub between a first position and a second position against a biasing force, and ii) pivotal about respective radial axes. A pitch linkage mechanism is operatively connected between each propeller blade and the main hub such that each propeller blade is pivotal about the respective radial axis from a first pitch orientation to a second pitch orientation together with movement of the propeller hub relative to the main hub from the first position to the second position responsive to air resistance in the second direction exceeding the biasing force.Type: ApplicationFiled: March 31, 2015Publication date: January 14, 2016Inventors: Andrew Kondor, Christopher Michael Rudnicki, Michael Anthony Rudnicki, Tyler J. Publication number: 20140290505Abstract: Waste objects, for example used oil barrels, are crushed using a device having a frame which supports first and second press plates so as to be movable towards one another in a crushing direction. A plurality of ball screw assemblies which are connected between the press plates each include a lead screw and a follower nut with roller elements in rolling engagement with the threads of the lead screw such that there is little or no static friction which must be overcome in order to release the press plate from the crushed object once the crushing operation is complete. A motor coupled to the ball screw assemblies applies a continuous torque throughout a full range of crushing movement of the second press plate relative to the first press plate to overcome the back pressure from the object being crushed.Type: ApplicationFiled: March 28, 2014Publication date: October 2, 2014Inventor: Michael Rudnicki
