    Abstract: The present disclosure relates to compositions and methods for reducing expression of MYC gene in a cell. In some embodiments, an expression repressor comprises a targeting moiety that binds a MYC promoter, anchor sequence, or super-enhancer. In some embodiments, the expression repressor comprises an effector moiety that represses transcription or methylates DNA. Systems comprising two expression repressors are also disclosed. The compositions can be used, for example, to treat cancers such as HCC or NSCLC.

    Abstract: Detecting analytes using proximity-induced tagmentation, strand invasion, restriction, or ligation is provided herein. In some examples, detecting an analyte includes coupling a donor recognition probe to a first portion of the analyte. The donor recognition probe includes a first recognition element specific to the first portion of the analyte, a first oligonucleotide corresponding to the first portion, and a transposase coupled to the first recognition element and the first oligonucleotide. An acceptor recognition probe is coupled to a second portion of the analyte. The acceptor recognition probe includes a second recognition element specific to the second portion of the analyte and a second oligonucleotide coupled to the second recognition element and corresponding to the second portion. The transposase is used to generate a reporter polynucleotide including the first and second oligonucleotides. The analyte is detected based on the reporter including comprising the first and second oligonucleotides.

    Abstract: Described are systems and assays that monitor presence and/or quantity of herpesviruses viral proteins. Embodiments offer accurate detection and quantification of viral proteins from all temporal classes of viral replication. Three exemplary assays provide specific detection of: herpes simplex vims type 1 (HSV1), human cytomegalovirus (HCMV), and Kaposi's sarcoma-associated herpesvirus (KSHV). These assays can be utilized in combination with drug treatments, genetic modifications, or other perturbations to assess the impact of the intervention on viral protein production.
