Abstract: The present disclosure provides a nucleic acid sequence for regulating the transcription of a nucleic acid fragment of interest, wherein the nucleic acid sequence comprises at least 2 copies of TetO-operator sequences capable of binding to a transactivator rtTA regulatable by tetracycline or a derivative thereof, and 1 copy of minimal promoter sequence containing a TATA box sequence, and at least 1 copy of a CuO-operator sequence capable of binding to a transcription repressor CymR regulatable by cumate. The present disclosure also provides a vector and a host cell containing the nucleic acid sequence, and a method for inducing the expression of a nucleic acid fragment of interest in a host cell.

Abstract: The disclosure provides an apparatus for processing a blood sample, a device for cells preparation and a method thereof. The disclosure can be used for blood separation, cell culture and preparation of cells. The apparatus mainly uses a weight sensor to have the weight and some liquid sensors. By optimizing the connection relationship of each device in the whole equipment, simplifying the operation process of the equipment, coordinating each step of the device, reducing the production cost, integrating the steps of PBMC cell separation and magnetic bead separation, the method makes the cell preparation process more intelligently automated, the operation simpler, the cell pollution reduced and the success rate of cell preparation improved. It has broad application prospects and huge market value.

The stationary part includes a cap, a socket connected to the cap, and a tube located within the cap and the socket. The rotatable part includes a centrifugal processing chamber, and a flow guide located within the centrifugal processing chamber. The stationary part and the rotatable part are connected by a plurality of rotary dynamic seals allowing rotation of the rotatable part relative to the stationary part. The cap includes a first port connected to an inlet, and a second port connected to an outlet. The centrifugal processing chamber includes an upper wall, a neck extending from the upper wall, a sidewall and a bottom wall tilting downwardly.
