    Abstract: Provided herein are methods and systems for the detection of spores in a sample which comprise permeabilizing a protein-based spore coat with a protein degrading agent comprising a non-ionic detergent and in particular with a specific mixture of various protein degrading agents comprising a non-ionic detergent to allow contact of spore nucleic acids with fluorescent reagents suitable for detection.
