    Abstract: The invention relates to mutant strains of the genus Sphingomonas which have a mutation in at least one gene encoding a protein involved in polyhydroxybutyrate (“PHB”) synthesis that allows the mutant strains to produce PHB-deficient Sphingans. The invention is also directed to a process for preparing a clarified Sphingan solution comprising heating aqueous Sphingan solution, in particular PUB-deficient Sphingan solution, to a clarification temperature of about 30° C. to about 70° C., and treating the solution with a clarification agent and enzymes. Certain of the Sphingomonas strains are diutan-producing strains that exhibit a dramatic improvement in productivity and yield due to a combination of certain genetic modifications that affect PHB and sphingan synthesis. Moreover, the sphingans produced from such strains have superior characteristics including improved filterability, clarity, and improved rheology-modifying characteristics. The sphingans provided are, thus, highly desirable in a variety of commercial and industrial uses, including personal care items, cement applications, and oilfield applications.

    Abstract: Mutational inactivation of proteins involved in para-cresol production in certain milk products results in improved taste and odor. The undesirable para-cresol forms over time as a result of enzymes produced by the bacterium that produces gellan gum. Since the gellan is typically used in a relatively unpurified form, the enzymes are added to the milk along with the gellan. Inactivation of the enzymes is a genetic means of eliminating the enzymes without requiring any additional purification or processing.

    Abstract: This invention provides a mutant Sphingomonas microorganism which produces a polysaccharide polymer comprising repeating tetramer units having a D-glucose:D-glucuronic acid:L-rhamnose ratio of about 2:1:1, wherein the D-glucose moieties are linked in a &bgr;-[1,4] configuration to the D-glucuronic acid moiety. One of the D-glucose moieties is linked to the L-rhamnose moiety in an &agr;-[1,3] configuration, the other glucose moiety is linked to the L-rhamnose moiety in a &bgr;-[1,4] configuration, wherein the polysaccharide polymer is substantially non-acetylated. This invention also provides the substantially non-acetylated polymer, as well as process for obtaining the substantially non-acetylated polymer.
