    Abstract: In the present invention, for test cells which are either stem cells whose state of differentiation is unknown or cells obtained from stem cells by differentiation induction, an LC-MS or GC-MS analysis is performed on culture supernatants collected from a culture dish of the test cells and a culture dish of control cells whose state of differentiation is known, and the state of differentiation of the test cells is assessed based on the amount, determined as a result of the aforementioned analysis, of at least one compound selected from the group of putrescine, kynurenine, cystathionine, ascorbic acid, riboflavin, pyruvic acid, serine, cysteine, threonic acid, citric acid, and orotic acid in both the culture supernatant of the test cells and the culture supernatant of the control cells.

    Abstract: A culture system has a culture bag for suspending and culturing pluripotent stem cells in a culture medium; a waste liquid container for storing used culture medium that is connected to the culture bag; a fresh medium container for storing a fresh culture medium that is connected to the culture bag; three-way stopcocks for switching flow channels from the culture bag to the waste liquid container or the fresh culture medium container, etc.; a trap portion for trapping the pluripotent stem cells in the culture medium in the flow toward the waste liquid container side; and a filter portion for subculturing that is formed in fourth and fifth flow channels in parallel to a first flow channel. The first flow channel connects the trap part to the culture bag, and has a mesh by which a cell mass of the pluripotent stem cells can be divided. A pump is used to pump the liquid in the individual flow channels.

    Abstract: The present invention provides a method of maintaining and amplifying pluripotent stem cells, including repeating the following steps: (i) suspension culturing pluripotent stem cells until cell aggregates have an average diameter of about 200-about 300 ?m, (ii) fragmenting the cell aggregates obtained by step (i) into cell aggregates having a uniform average diameter of about 80-about 120 ?m. In step (i), a suitable viscosity is conferred to the medium to prevent movement of floating cell aggregates and adhesion and fusion of cell aggregates. In step (ii), the cell aggregates are mechanically fragmented into smaller uniform cell aggregates by passing the cell suspension through a mesh.
