  • Publication number: 20230304996
Abstract: Disclosed is a microfluidic device implanted in an ex-vivo skin explant to control perfusion, diffusion and collection of molecules over long periods. Also disclosed is a method for assessing permeation or infusion of a biomarker of interest through the skin and to a method for detecting and/or quantifying a biomarker of interest contained in the liquid secreted by the ex-vivo skin explant.

  • Publication number: 20200400652
Abstract: Disclosed is an ex vivo model for subcutaneous injection and aims at providing an in vitro method including the steps of i) immersing a skin explant in a solidifiable liquid matrix such that the upper face of the epidermis is not covered, which matrix is itself contained in a cell culture insert, the bottom of which consists of a porous membrane, and (ii) solidifying this matrix so as to trap the immersed portion of this skin explant, wherein the upper face of the epidermis is not covered, and adhering this same matrix to the side walls and the porous membrane of the insert, wherein the skin explant includes a thickness of at least 5 mm of hypodermis.

  • Publication number: 20200347429
Abstract: Disclosed is an ex-vivo model of inflamed skin, an in-vitro method for obtaining the same and uses thereof for screening anti-inflammatory compounds. The method includes injecting, into the dermis of a healthy skin biopsy previously taken from a mammal, a composition including an effective amount for activating dermal resident T cells of an anti-CD3 antibody and an anti-CD28 antibody; and b) incubating the injected skin biopsy obtained in step a) in the presence of a composition including an effective amount, for obtaining the polarization of the T cells activated in step a) into LTh1 and/or LTh17 and the synthesis of inflammation markers, of at least one mixture of IL-1?, IL-23, and TGF-?.
