  • Patent number: 8512959
Abstract: A method or platform for monoclonal antibody based biomarker discovery is disclosed. The method according to the invention provides for the integration of analyte collection, hybridoma screening and nanovolume integrated mass spectrometry (NVIMS) to achieve a robust screening system that is capable, for example, of cutting 4-6 years off of the classical biomarker discovery and development process. The invention provides a platform for the rapid, high-throughput production, isolation and characterization of, e.g., disease specific biomarkers together with highly specific monoclonal antibodies. The method of the invention has a variety of applications such as, but not limited to, drug testing, biohazard applications, ecological applications, physiological applications and/or pathology screening applications. The method of the invention is also capable of being performed or used as or with a high-throughput screening process or system of the invention.
    Type: Grant
    Filed: February 9, 2005
Date of Patent: August 20, 2013
    Assignee: Northwestern University
Inventors: Laszlo Takacs, Andras Guttman, William S. Hancock, Barry L. Karger, Manuel Duval, Patrick Berna

  • Publication number: 20040102615
Abstract: The invention concerns a method for isolating and purifying a protein of interest, in particular from a complex medium such as a plant extract. Said method is characterized in that it compromises a step whereby a complex medium, comprising the solution containing the protein of interest to be purified and a solid support capable of enabling it absorption, is brought in the presence of an agent capable of causing said protein to precipitate in soluble form. The protein of interest is thus partly aggregated and absorbed on the solid support without substantial formation of macro-aggregates in the solution capable of spontaneous elutriation.

The invention also relates to pharmaceutical compositions comprising a recombinant gastric lipase and methods of use.

  • Publication number: 20030100019
Abstract: The present invention describes a new artificial supported membrane, as well as methods of preparation of such membrane or of reconstitution of cell membranes from their constituent proteins and lipids. Such membrane comprises a lipid bilayer attached to a support by phospholipid tethers (P) creating a space between the bilayer and the support, as well as one or more ligands (L) specific of a protein, covalently bound to the support and exposed in said space. The invention equally describes the uses of such membranes which allow the purification and/or reversible capture of membrane proteins or yet the screening of compounds that interact with some of these proteins. A membrane according to the invention may further serve to evaluate the toxicity or, conversely, the therapeutic effect of test compounds.
