    Abstract: A method of reproducibly generating dendritic cells is provided. Blood mononuclear cells is loaded into a cell culture container containing microcarrier beads therein. Tissue culture comprising the cells loaded in the container is incubated for a predetermined period. Nonadherent cells and cells adhered to the beads are separated. The method may further include preparing dendritic cell culture medium and transferring the dendritic cell culture medium to the container after the cells which are adhered to the beads are separated from the nonadherent cells. The tissue culture incubated for the predetermined time period may be washed to remove nonadherent cells. The beads may be allowed to settle and supernatant is expressed off. The container may comprise a gas permeable cell culture bag.
