    Abstract: A rapamycin (RAPA) composition and a preparation method thereof are provided. The RAPA composition includes the following active ingredients in parts by weight: RAPA: 1 to 10 parts; polymer carrier: 0.5 to 20 parts; and lymphatic target: 0.1 to 1 part. The lymphatic target is at least one selected from the group consisting of sodium hyaluronate (SH), an aptamer, and an antibody. The preparation method includes: preparing an organic phase solution by adding RAPA to an organic phase solvent to obtain the organic phase solution; preparing an emulsion by adding a polymer carrier to an aqueous phase solvent and adding the organic phase solution dropwise to the aqueous phase solvent to obtain the emulsion; and conducting homogenization by adding a lymphatic target to the emulsion, mixing, and homogenizing to obtain the RAPA composition. The RAPA composition can target a lymphatic system to treat atherosclerosis (AS) and related cardiovascular diseases (CVDs).

    Abstract: A rapamycin (RAPA) liposome formulation and a preparation method thereof are provided. The RAPA formulation includes (in parts by weight): RAPA: 1 to 100 parts; phospholipid: 1 to 2,000 parts; and stabilizer: 0.01 to 100 parts. The preparation method includes: mixing and dissolving RAPA, a phospholipid, and a stabilizer in an organic phase solvent to obtain an organic phase mixed solution; preparing an initial emulsion solution by adding the organic phase mixed solution dropwise to an aqueous phase solvent, and stirring at room temperature to obtain the initial emulsion solution; and conducting lyophilization by homogenizing the initial emulsion solution, adding a lyophilization protective agent, mixing, and filtering a resulting mixture through a microporous filter membrane for sterilization to obtain the RAPA formulation of a liposome-lyophilized powder.

    Abstract: A rapamycin (RAPA) self-microemulsifying injection includes a drug stock solution and a diluent. The drug stock solution includes a drug solvent, an antioxidant, and RAPA, and the diluent includes a surfactant and water. RAPA is dissolved in the drug solvent. After the stock solution is diluted with the diluent, the solution is self-assembled into a microemulsion under the action of a surfactant, and RAPA is encapsulated in the microemulsion, which improves the solubility and stability of RAPA.
