    Abstract: A chimeric antigen receptor (CAR) that binds CD117 (anti-CD117 CAR), wherein the anti-CD117 CAR comprises: (a) an ectodomain that binds CD 117; (b) a transmembrane domain; and (c) an endodomain comprising a costimulatory domain and a CD3? signaling domain. The ectodomain comprises a single chain variable fragment (scFv) that binds CD 117 (anti-CD117 scFv), or comprises a single domain antibody fragment such as a VHH fragment. Also provided herein are genetically engineered T cells expressing such anti-CD117 CAR, which can be used for inhibiting CD117+ cells such as cancer cells.
