    Abstract: The present disclosure provides recombinant adeno-associated virus virions with variant capsid protein, where the recombinant AAV (rAAV) virions exhibit one or more of increased ability to cross neuronal cell barriers, increased infectivity of a neural stem cell, increased infectivity of a neuronal cell, and reduced susceptibility to antibody neutralization, compared to a control AAV, and where the rAAV virions comprise a heterologous nucleic acid. The present disclosure provides methods of delivering a gene product to a neural stem cell or a neuronal cell in an individual. The present disclosure also provides methods of modifying a target nucleic acid present in a neural stem cell or neuronal cell.

Methods include cell-containing liquid droplets with oligonucleotide-containing liquid droplets, hybridizing oligonucleotides to target nucleic acids from cells, extending the hybridized oligonucleotides on the target nucleic acids into cell identifier sequences on the target nucleic acids, and thereby identifying the type of cells initially present. The methods can be implemented in a high-throughput manner in a microfluidic system.

    Abstract: The present invention generally relates to Magnetic Resonance Imaging (MRI) analysis of the location, presence, and/or quantity of a particular molecule or material. In some embodiments, the invention relates to compositions and methods for determination of an analyte, wherein a compositions may produce an MRI signal that can be affected by the presence of a particular composition (e.g., analyte). In a particular embodiment, the composition comprises a protein. The invention also provides related nucleic acid molecules, polypeptides, and fragments thereof. Compositions of the invention may exhibit reduced toxicity and may be readily delivered in vivo. Various embodiments of the invention may be useful as sensors, diagnostic tools, and the like.
