    Abstract: A method for preparing a porous-three-dimensional scaffold good for tissue engineering is described. Sericin forms a three-dimensional scaffold with PVA after freeze-drying having glycerin as a plasticizer and genipin as natural crosslinking agent to help making a strong and stable matrix. Adding glycerin into scaffold gives good uniformity and porosity. Smaller pore sizes and better uniformity are obtained as the concentration of genipin in the scaffold increases. Glycerin retains a high moisture content to allow the presence of water molecule in the matrix structure. Adding genipin results in a higher degree of crosslinking within the scaffold. Crosslinking using genipin is most beneficial in preparing scaffold possesses the best biological and physical properties for wound healing. The present invention describes method for preparing crosslinked matrix whose composition can be appropriately tuned to obtain matrix with desirable characteristics for biological applications.
