    Abstract: A device is disclosed for conducting an assay method. The device comprises a housing, means enclosed in the housing for capturing a member of a specific binding pair in a zone and for allowing liquid to be transported by capillary action away from the zone, one or more self-contained liquid reagents enclosed in the housing for conducting an assay method for the determination of an analyte in the sample, and means in the housing for introducing the sample into the device. Preferably, the self-contained reagents are liquid reagents which are contained in a breakable container. The device of the invention finds use in assay methods for the determination of an analyte in a sample suspected of containing the analyte. Kits for conducting an assay method are also disclosed.

    Abstract: Polyiodothyronine analog conjugates to glucose-6-phosphate dehydrogenase (G6PDH) are provided which find use in the determination of polyiodothyronine compounds, particularly thyroxine, normally in physiological fluids, such as serum. The G6PDH enzyme activity can be substantially reduced when bound to antibodies specific to polyiodothyronine as compared to the free or unbound polyiodothyronine analog enzyme conjugates. The polyiodothyronine analog is conjugated to the G6PDH by a linking group having a chain one atom in length.

    Abstract: A method is disclosed for preparing a sample suspected of containing digoxin for determination of digoxin in the sample during an assay. The method comprises contacting the sample with .beta.-cyclodextrin in an amount and under conditions sufficient to allow a substantial portion of digoxin in the sample to bind to the .beta.-cyclodextrin. The .beta.-cyclodextrin with bound digoxin is separated from at least one other component of the medium. Next, the digoxin is released from the .beta.-cyclodextrin to provide a sample containing digoxin which may be analyzed by any of a number of assay techniques.

    Abstract: A method is disclosed for determining an analyte in a sample suspected of containing the analyte. The analyte is a member of a specific binding pair (sbp) consisting of ligand and complementary receptor. The method comprises combining in an aqueous medium (1) said sample, (2) a first reagent comprising a non-dispersed surface to which is bound an sbp member that becomes bound to the surface in relation to the amount of analyte in the sample. The volume of the aqueous medium containing the above reagents is sufficiently large to allow complete immersion of the first reagent therein and determination of the analyte but not so large to result in substantial interference in the determination upon addition of a second reagent reactive with the conjugate of enzyme and sbp member and capable of generating a signal. The second reagent is present in a medium of sufficient volume to substantially increase the volume of the liquid medium and reduce interference in the determination.

    Abstract: A method and device for determining the presence of an analyte in a sample suspected of containing the analyte is disclosed. The method involves contacting a test solution containing the sample and a first member of a specific binding pair with an end portion of a strip of bibulous material capable of being traversed by the test solution through capillary action. The first member of a specific binding pair is capable of binding the analyte. The strip contains a second member of a specific binding pair integral therewith for concentrating and non-diffusively binding the first sbp member at a small situs on the strip separated from the end portion of the strip. The detectible signal is produced in relation to the presence of the analyte in the test solution. The test solution passes through the situs as the test solution traverses the bibulous material.
