  • Publication number: 20250250613
Abstract: The subject invention pertains to a method for identifying G-quadruplexes (G4s) in a cell using a G4 fluorescent probe, 2-(2-hydroxy-6-methoxy-3-propionylphenyl)quinazolin-4(3H)-one (HMPQ), which exhibits specific binding affinity to G-quadruplexes (G4s), illuminating these non-canonical nucleic acid structures without altering their conformation. G4s are involved in chromatin organisation, gene regulation, genome stability, and appear to contribute to cancer growth and progression. By utilizing HMPQ's unique fluorescence mechanism through excited-state intramolecular proton transfer, the method allows label-free precise detection of G4 structures in the nucleus of a cell.

  • Publication number: 20250250574
Abstract: The present invention provides an siRNA or a pharmaceutically acceptable salt thereof for inhibiting TMPRSS6 expression in human cells, as well as appropriate modifications of the siRNA to enhance target silencing efficiency and minimize off-target activity. The invention further provides biological agent or pharmaceutical compositions containing the foregoing siRNA for inhibiting TMPRSS6 expression. Through a series of in vitro and in vivo experiments, the present invention has identified siRNA sequences with potent biological activity in suppressing TMPRSS6 expression, demonstrating higher efficacy than TMPRSS6-HCM-9, which is currently the most advanced siRNA in clinical development. These findings support the potential clinical application of the invention in treating disorders associated with dysregulated TMPRSS6 expression or diseases related to iron excess or iron overload, demonstrating significant therapeutic potential.
