    Abstract: A method for identifying biologically significant peptide-DNA binding interactions and sequence-specific DNA-binding peptides in vivo using combinatorial oligonucleotide libraries is disclosed. Target DNA sequences include promoter sequences, 5' and 3' regulatory sequences, and exon and intron regulatory sequences. Preferably, a yeast combinatorial library is employed. Peptides that bind to specific DNA sequences, identified by the above method, are disclosed. Also disclosed is a method for identifying nontoxic compounds that inhibit gene expression in host cells in vivo.
