  • Publication number: 20230399672
Abstract: The present invention in various aspects and embodiments provides engineered bacterial strains having a modified genome that provides advantages in maintenance and biosynthetic processes when cultured at large scale. The present invention further provides methods for large-scale fermentation using the bacterial strains. In various embodiments, the strain engineering yields a reduction in cellular responses to micro-environmental stimuli imposed in large scale bioreactors.

  • Publication number: 20190144815
Abstract: The present invention relates to recombinant Escherichia coli (E. coli) host cells comprising, in relation to wild-type cells, at least one mutation selected from the group consisting of deletion of the gene relA (?relA); amino acid substitutions R290E and K292D in the protein guanosine-3?,5?-bis pyrophosphate 3?-pyrophosphohydrolase (bifunctional (p)ppGpp synthetase II; SpoT) (spo T[R290E;K292D]); and amino acid substitution G267C in the protein pyruvate dehydrogenase subunit E1 (AceE) (aceE[G267C]). Said recombinant host cells are characterized by increased sugar uptake rates that lead to increased productivity when using said cells for the production of biosynthetic products. The present invention further relates to respective methods for the biosynthetic production of a product of interest using said host cells.

  • Publication number: 20060191848
Abstract: The present invention relates to a process for integrated removal of one or more organic substances present in an aqueous bioprocess mixture, which comprise at least one positively charged and/or chargeable nitrogen-containing group, by means of reactive extraction in at least one step, in which process at least one liquid-liquid centrifuge is used for said reactive extraction and said organic substances are re-extracted from the extractant into an aqueous phase. More specifically, the bioprocess mixture directed to the liquid-liquid centrifuges is rendered cell-free and protein-free, before being directed into the first centrifuge. Furthermore, the invention relates to such a process in which an aqueous bioprocess mixture is continuously removed from a bioreactor, led, with an extractant, into a liquid-liquid centrifuge, extracted by means of said extractant, with an organic phase being obtained which contains the substance to be extracted from the fermentation mixture.

  • Publication number: 20050227333
Abstract: The invention relates to a process for the production of an aromatic amino acid metabolite or derivative thereof by aerobic fermentation of Escherichia coli, which fermentation comprises a growth and a production phase and in which fermentation glucose and L-tyrosine are controlled, wherein during at least part of the production phase, the glucose concentration in the fermentation medium is controlled within the range of 1-20 g/L and the L-tyrosine concentration in the fermentation medium is controlled below 36 mg/L.

  • Patent number: 6793825
Abstract: The present invention relates to a process for separating from an aqueous mixture one or more organic substances containing at least one positively charged and/or chargeable nitrogenous group by means of extraction via at least one porous membrane, wherein use is made of an extraction agent which contains at least partially relatively long-chain organic compounds and at least one liquid cation exchanger, and of a membrane that is wettable by either the aqueous mixture or by the extraction agent.

  • Publication number: 20030224495
Abstract: In the production of genetically-modified, non-pathogenic organisms particularly an improved production and isolation of trans-dihydroxycyclohexadiene carboxylic acids and/or derivatives thereof, the synthesis of the transhydroxy cyclohexadiene carboxylic acids is performed in an enantiomer-pure form without detectable impurities with chorismate by cultivation of at least one genetically altered non-pathogenic organism which has an aromate embolism and has, in comparison with a corresponding, genetically not altered organism, an increased activity of the isochorismate synthase (EntC) and isochorismatase (EntB) and an activity of the 2,3 dihydroxybenzoate synthase (EntA) which is close to zero, or an increased activity of the isochorismatase (EntB) and a reduced activity of the isochorismate synthase (EntC) and an activity of the 2,3-dihydroxybenzoat-synthase (entA) of close to zero.
