  • Publication number: 20100311072
Abstract: The subject matter disclosed and claimed herein concerns measuring the binding affinity of glucokinase (“GK”) using a fluorescence polarization (“FP”) assay. The FP method includes use of modified GK ligands bound to a fluorescent label. Binding affinity is determined by measuring displacement of fluorescent ligand by the known or suspected GK ligands. The subject matter disclosed and claimed herein provides a robust high-throughput FP assay for the determination of binding affinity of ligands to glucokinase. The FP binding assay displayed both glucose and nucleotide dependence, and a useful dynamic range. The binding IC50 data correlated well with GK activation EC50 data.

  • Patent number: 7384757
Abstract: The present invention relates generally to enzyme assays and more particularly to a rapid, sensitive, reliable and robust homogeneous assay for acetyl CoA carboxylase activity comprising a coupled enzyme assay in a scintillation proximity format suitable for high-throughput screening. In one aspect, the assay couples ACC and FAS and the product, palmitic acid is then detected by scintillation counting. The invention also relates to the identification of modulators of ACC activity.

  • Patent number: 5635503
Abstract: A series of non-peptidergic antagonists of NPY have been synthesized and are comprised of piperazine and homopiperazine derivatives of 4-phenyl-1,4-dihydropyridines of Formula (I). ##STR1## As antagonists of NPY-induced feeding behavior, these compounds are expected to act as effective anorexiant agents in promoting weight loss and treating eating disorders.
